Building Muscle After 40: What Actually Changes (and What Doesn't)

The muscle-building signal doesn't disappear at 40. What changes is anabolic efficiency: how much protein it takes to trigger growth, and how often. Here's the mechanism and the numbers.

"Is it even still possible to build muscle at my age?" The honest answer is yes. The biology that makes muscle grow doesn't switch off at 40. What changes is something less dramatic and more fixable: your body gets less efficient at converting the protein you eat into new muscle tissue. That's a numbers problem, not a verdict.

This piece covers what actually changes after 40 (anabolic efficiency), what doesn't (the growth mechanism itself), and the specific protein targets, daily and per-meal, that compensate for the difference.

The muscle-building signal doesn't disappear at 40

Muscle growth runs on one core mechanism. Mechanical tension from resistance training activates a signaling pathway (mTOR) that ramps up muscle protein synthesis (MPS), literally building new contractile tissue. That pathway isn't age-gated. It responds to a heavy set of squats at 45 the same way it responds at 25.

What does shift slightly is the loading strategy that triggers it best. Schoenfeld & Grgic (2021), re-examining the loading literature, found comparable hypertrophy from roughly 30% of 1RM up through heavy loads, about 5–30 reps per set, as long as sets are taken close to failure. That range gives adults managing joint wear more room to train hard without maxing out on heavy singles. The stimulus works the same; you just have more valid ways to apply it.

So if the mechanism is intact, why does "can I still build muscle after 40" feel like a real question? Because a second, less-discussed variable changes with age, and it's the one that actually explains the gap between "the mechanism works" and "results feel harder to get."

What actually changes: anabolic resistance

The mechanism doesn't degrade. The efficiency of triggering it does. This is called anabolic resistance: older muscle needs a bigger stimulus (more protein, more mechanical tension) to produce the same protein-synthesis response that younger muscle gets from a smaller one.

Wall et al. (2015), pooling data from six stable-isotope tracer studies across roughly 180 participants, found post-meal muscle protein synthesis rates were 16% lower in older adults than in young adults eating the same meal. The gap was even starker in how much each group's MPS rate rose from resting to fed: young adults saw about a 75% jump from a meal, while older adults saw only about a 21% jump.

The muscle isn't refusing to grow. It's responding to the same input with roughly a third of the sensitivity. The practical fix is proportional: give it a bigger input.

The per-meal protein threshold

Anabolic resistance isn't just about the day's total protein. It's about whether each individual meal clears a threshold high enough to trigger a response at all.

Katsanos et al. (2005) tested this directly, feeding elderly and young adults essential-amino-acid mixtures at different leucine concentrations. Older adults showed essentially no muscle-protein-synthesis response to 1.7g of leucine, roughly what you'd get from 15g of whey protein, about a half-scoop of a typical protein shake. Raise that to 2.8g of leucine, roughly 25–30g of whey protein, closer to a full scoop plus, and the response became robust, comparable to what younger adults get from meals with less leucine.

That's the practical number: a meal light enough to "count" toward a daily protein goal on paper can still be too small to trigger meaningful muscle protein synthesis if you're over 40. Three or four meals a day each carrying 25–30g of complete protein does more for actual muscle-building than the same total protein loaded mostly into one or two meals.

How much protein, total

Per-meal thresholds only matter within a sensible daily target. Bauer et al. (2013), the PROT-AGE Study Group's expert consensus position paper, recommends 1.0–1.2g of protein per kilogram of body weight per day for older adults generally, with ≥1.2g/kg recommended specifically for those who are exercising and physically active, which describes anyone running a strength program in this age range.

Worth flagging directly: PROT-AGE's subject population is adults over 65, a decade or two past most of this article's readers. The mechanism it's describing, anabolic resistance, starts earlier and builds gradually rather than switching on at 65. So the honest read isn't "this number applies to you exactly." It's closer to "the safety margin from erring toward the higher end of this range costs little and buys real insurance as you move through your 40s and 50s."

That range sits below the daily target already established for adults in this specific age bracket. Strength training in your 40s covers the fuller picture: Morton et al. (2018), a meta-analysis of 49 studies and 1,863 participants, found strength and lean-mass gains from protein plateaued around 1.6g per kilogram of body weight per day, roughly 0.7g per pound. That's the number to actually target; PROT-AGE's lower range is the floor for general lean-mass maintenance, not the ceiling for someone actively training for growth.

A sample day: 3–4 meals, protein distributed to clear the threshold

Here's what hitting both the daily target (~1.6g/kg) and the per-meal threshold (~25–30g) looks like for a 175lb (79kg) adult targeting roughly 125g of daily protein:

Every meal clears the ~25–30g threshold the Katsanos research points to, and the daily total lands within the range that supports lean-mass gains at this age. The exact foods don't matter. Per-meal size and daily total are the two constraints that do.

What doesn't matter as much as people think

Testosterone decline gets blamed for a lot of this, and it's real but overstated as an explanation. Testosterone falls gradually, roughly 1–2% per year after 30 based on longitudinal data, and by 45 that can mean 15–30% below peak. It influences the rate of muscle protein synthesis and recovery speed, not whether the stimulus-response relationship works at all. Adults with lower testosterone still build muscle in response to training; the anabolic-resistance mechanism above explains far more of the "why does this feel harder" gap than hormone levels do.

The takeaway: this is an inputs problem, not a motivation problem

Nothing about turning 40 shuts off the biology that builds muscle. What it does is raise the bar on the inputs: more protein per meal to clear the response threshold, a daily total in the 1.6g/kg range rather than the 0.8g/kg most general dietary guidelines suggest, and training that hits each muscle group with enough frequency and load to keep asking for growth. The actual program supplies the training side. This piece supplies the eating side. Neither works without the other, and neither works without running it long enough to matter.
